The present invention relates to a method for increasing recombinant human acid α-glucosidase productivity, and more particularly, to a composition for increasing productivity and a method for increasing productivity of recombinant human acid α-glucosidase including a self-extinguishing inhibitor. The autodigestion inhibitor according to the present invention can suppress autodigestion caused by sugar depletion in a transformed rice cell culture environment, thereby enhancing the survival of rice cells and increasing the productivity of recombinant human acid α-glucosidase. In particular, even in transgenic rice cells derived from glycoengineered rice cells, which are more sensitive to sugar depletion environments, the productivity of recombinant human acid α-glucosidase can be increased, which can be useful for producing recombinant human acid α-glucosidase.
