This is achieved by the fact that in the method of replacing mitochondria and cytoplasm in oocytes having a pathogenic mutation in mtDNA or cytoplasmic insufficiency, genetic testing is performed according to the invention to determine mtDNA from blood and / or fibroblasts of the skin and / or urine of a recipient mtDNA carrier having pathogenic mutation, the metaphase II spindle is isolated from the recipient oocyte of the mtDNA carrier having a pathogenic mutation or pathology in the cytoplasm, the metaphase II spindle is removed from the healthy donor oocyte, the metaphase II spindle from the recipient oocyte is transferred to the healthy donor's oocyte cell, the patient is injected recipient oocyte with a spindle transferred to obtain an embryo, the embryo is cultured with the formation of a blastocyst and pregnancy is established.
