The invention relates to a microstructured substrate which can be used to perform surface plasmon resonance measurements, having applications in the field of biosensors, and to a process for manufacturing the same. The claimed substrate is made up of a glass substrate, an optically transparent metallic film, a rarefied network of polymeric fibres, and an additional optically transparent metallic film. The claimed process consists in applying an oxygen plasma cleaning/activation treatment onto a glass plate, coating it with an optically transparent metallic thin film, applying a second oxygen plasma cleaning/activating treatment onto the metal-coated plate, deposition, on the metal-coated plate, of a rarefied network of polymeric fibres by electro-spinning, application of a new oxygen plasma cleaning/activation treatment of the metal-coated and fiber-coated plate, and deposition of an additional metallic thin film, the successive plasma treatments and the deposition of the polymeric fibre network improving the adhesion of the metal layer, the mechanical strength and the operating life of the whole assembly, while the electrospun fibres can improve the plasmon response.
