The well-known drug colchicine with various approved therapeutic properties in systemic and topical diseases could be chosen to be formulated in a nanofiberous based drug delivery system that it may enhance the potency and also it reduces the systemic toxicity in local administrations. In this invention, colchicine loaded chitosan based nanofiberous system was formulated, optimized, fully characterized, and evaluated in ex vivo. The optimizations was analyzed via AFM and SEM based on the morphology, topology and the diameters of the nanofibers. In addition, FTIR, XRD, STA, water angle measurement, tensile evaluation, and ex vivo skin permeation were checked. The nanofibers provided a significant colchicine deposition and a notable first order release kinetic behavior. This makes it a formulation to be administered for topical drug delivery or local reservoir for neoadjuvant therapy applying the anticancer property of colchicine, and also as a transdermal colchicine patches.
