The present invention relates to oligonucleotide-coded libraries and methods of tagging the libraries. In particular, the methods and oligonucleotides can be prepared by chemically ligating one or more 2'-substituted nucleotides, such as 2'-O-methyl or 2'-fluoro nucleotides, and other conditions or reagents that enhance enzymatic ligation. It may include one or more supporting chemical functional groups.
