Mutamba plant (Guazuma ulmifolia Lamk.) is one of herbal plant that has udbeen used by a lot of people in Indonesia. The main medicinal used of this plant is udfor slimming body. Secondary metabolites ingredients of mutamba plant which udare medicinal useful can be produced rapidly by in vitro callus induction. This udresearch was a callus induction of mutamba plant in various media composition udunder light and dark condition. udThis research was aimed to study the comparison of callus induction in udvarious media composition under light and dark condition. The research was udconducted in Plant Physiology and Biotechnology Laboratory of Agriculture udFaculty Sebelas Maret University from December 2007 until March 2008, used udRandom Complete Design with single factor consist of 7 treatments repeated 3 udtimes. The treatments were Gamborg (B5) + sucrose 2% + 2,4D 1 mg/l (light); udGamborg (B5) + sucrose 2% + 2,4D 1 mg/l (dark); Gamborg (B5) + sucrose 3% + ud2,4D 2 mg/l (light); Gamborg (B5) + sucrose 3% + 2,4D 2 mg/l (dark); MS + udsucrose 8% + 2,4D 1 mg/l (light); MS + sucrose 8% + 2,4D 1 mg/l (dark) and MS ud+ 2,4-D 0,6 mg/l + 1 mg/l kinetin (control) udThe result observation data of callus initiation time, callus volume, fresh udweight callus and dry weight callus variables were analyzed with variance test that udseparated in comparison by Contrast Test in level 5%. Contrast test among the udtreatment were : control with all treatments; B5 medium with MS medium; light udcondition with dark condition; 1 mg/l 2,4-D with 2 mg/l 2,4-D; 2 mg/l 2,4-D light udcondition with 2,4-D dark condition; 2% and 3% sucrose with 8% sucrose. Data udof texture and colour callus variables were analyzed descriptively. Callus volume udand fresh callus weight variable were also analyzed using correlation test. The result showed that callus growth in 2,4-D with same concentration in udlight condition was better than in dark condition. The use of 1 mg/l kinetin and 0,6 udmg/l in MS media was not different in callus formation compare with media udwhich only used 2,4-D. The treatment which showed the best callus growth was udGamborg (B5) + sucrose 3% + 2,4D 2 mg/l (light). ud
