Two different molecular tools for theuddiagnosis of the cereal and legume root-lesionudnematode Pratylenchus thornei were developed.udA randomly amplified DNA (RAPD) fragmentudspecific to P. thornei was identified. Afterudsequencing the fragment, longer primers wereuddesigned that complement the terminal sequencesudof the RAPD fragment, and this pair of specificudprimers was used to amplify the sequence-characterizedudamplified region (SCAR). Using theuddeveloped pair of SCAR primers, the SCARudfragment specific to P. thornei was easily amplifiedudwith DNA extracts obtained from differentudlife stages of the nematode. The described SCARPCR-udbased assay has the potential to be optimizedudfor routine practical diagnostic tests. Inudaddition, the use of a species-specific satelliteudDNA sequence to distinguish P. thornei fromudother Pratylenchus spp. is discussed.
