Acute and Repeated Dose Toxicity Studies on Nsc 94100 Following Daily Oral Administration to Dogs and Monkeys and Acute Toxicity Following Oral Administration to Mice and Monkeys

Compound NSC 94100, D-Mannitol, 1,6-dibromo- 1,6-dideoxy- was studied following daily oral administration in beagle dogs in dosages of 200 mg/kg x 6, 100 mg/kg x 5, 50 mg/kg x 10, 35 mg/kg x 12, 25 mg/kg x 14, and 13 mg/kg x 90, the dosages being administered in gelatin capsules and in Rhesus monkeys at daily dosages of 150 mg/kg x 8, 75 mg/kg x 8, 38 mg/kg x 9, 19 mg/kg x 21 and 57, and 10 mg/kg x 200. The drug was administered to the monkeys by gavage as a suspension in water. The tolerable dose in dogs was 13 mg/kg/day x 90 and the tolerable dose in the monkey was 10 mg/kg x 200. A single dose of 600 mg/kg P.O. was administered to a single dog. The oral LD50 was determined in mice and rats and range finding toxicity tests were done following I.P. administration in mice.
