Factors Related to Survival and Function of Bone Marrow Transplants in Rats

Efforts were made to test for immunoglobulin allotype differences in rats of the F344, Buf, WF, BN, Lewis and ACI strains. Antisera was prepared in the ACI strain by immunization with suspensions of proteus mirabilis coated with BN antibody. ACI animals produced a sera that gave a precipitation reaction in gel of identity with sera from all of the other rat strains. This component on immunoelectrophoresis migrated as an immunoglobulin. The various strains were examined further for hemoglobin differences. Hemoglobin from at least 10 members of each strain was electrophoresed on cellulose acetate. The various strains were found to be homogenous for their hemoglobin patterns as described in our previous report. Six components were identified. In brief, 3 patterns are discernible: WF, F344 type; BN, LBNF sub 1 type; and Lewis, Buf, ACI type. Further clarification of histocompatibility relationships between strains was investigated by skin grafting combined with Cyclophosphamide treatment. In addition, graft-versus-host assays were carried out in the BN strain. The results of the latter study indicate that the ACI and Buf strains are more closely related to BN than the other 3 rat strains. Lewis rats given a lethal dose of Busulfan, together with a non-lethal dose of Cyclophosphamide, were protected from drug lethality by allogeneic marrow infusions. Representative surviving animals at six weeks were cellular chimeras. A synergistic action of tee two drugs is proposed since neither drug given alone in the doses employed will condition the animal for allogeneic marrow transplants. (Author)
