These studies were initiated to determine whether the administration of saline in volume doses equivalent to 500 IU/kg of L-Asparaginase, B.I.D. x 5, according to the protocol for enzyme treated animals would result in (1) anemia, (2) leucocytosis, (3) alterations in BUN, (4) pyrogenic effect, (5) hepatotoxicity, (6) colitis and (7) weight loss. These parameters included the major changes noted following L-Asparaginase administration to Rhesus monkeys and some were noted in the beagle dog. In the beagle dog the administration of saline did not result in any overt toxic symptoms, anemia, leucocytosis, hepatotoxicity or colitis. In the Rhesus monkey the two major changes noted were anemia and leucocytosis following the first dose of saline. It is obvious from these results that the anemia noted in the Asparaginase treated animals was due to the multiple bleedings during the course of the study, and the leucocytosis noted in these animals is probably associated with stress. No biochemical or pathologic evidence of hepatotoxicity or colitis was noted in the saline treated monkeys and they showed no weight loss or any other overt toxic symptoms. (Author)
