Establishing a Primate Pre-Clinical Test Center for Immune Suppression Study (Bone Marrow Transplantation Studies)

Two goals were achieved, (a) the development of lymphocyte alloantigen typing in Rhesus monkeys and (b) the establishment of a preclinical bone marrow transplantation model in the Rhesus monkey. The lymphocytes of 42 unrelated Rhesus monkeys have been typed by trypan blue cytotoxicity with sera from 17 Rhesus monkeys. Four antigens have been tentatively identified and studies on their inheritance are beginning. Fifty percent of all multiparous (three or more pregnancies) Rhesus sera tested (13 of 26) were found to contain antibodies cytotoxic to Rhesus lymphocytes. A bone marrow transplantation model was established using lethal radiation to create marrow aplasia. Autologous marrow transplantation was successful in three of four attempts. The unsuccessful attempt died of a pulmonary embolus before evidence of graft take could be firmly established. Three monkeys were given mismatched allogeneic marrow and all died of acute graft-versus-host disease and had other evidence of allograft take. (Author)
