NATURE OF AND CARCINOGENIC POTENTIAL OF BETA-PROPIOLACTONE (BPL) REACTION PRODUCTS.

BPL was reacted with various common biological material and the resulting products were separated by various chromatographic techniques. Changes in the chromatograms did occur,demonstrating the high reactivity of BPL with biological material. Rf values for these various end products are given. The only reaction product positively identified was 3-chloropropionic acid, a resulting product of BPL in saline. Each of 13groups of 50 weanling CAF-1mice of both sexes (650total) received 24weekly subcutaneous injections of 0.1ml of one of the following: sterile saline,sterile peanut oil,or one of the 11 BPL reaction products. These reaction products were obtained by treating solutions of peanut oil,DNA,RNA,saline,plasma, (RNA +lecithin +albumin +DNA +heparin),cysteine,guanine,Salmonella, (adenine +pyrimidines) and adenovirus 12with 0.00278molar BPL. Where possible,the biological products were diluted to a 0.00278 M strength. All mice were continued on study for 18months or until death,at which time necropsy was performed. Gross necropsy findings were essentially similar in all groups except in the 25female weanling mice given BPL +peanut oil. Nineteen of these females had subcutaneous sarcomas compared to only three in the group receiving peanut oil alone,and none in those receiving saline. In comparable groups of male mice,the number with sarcomas were: BPL +peanut oil - 3/25,peanut oil - 1/25,and saline - 0/25. Attempts to transplant the sarcomas from two female mice were both successful. The group of nice given BPL +adenovirus 12had the highest incidence of lung adenomas (23/50) compared to 15/50in the group receiving saline. The presence of live residual adenovirus in the inoculum may have had some effect on these results.
