Polyvinylpyrrolidone (PVP) was evaluated for acute toxic effect by rapid intravenous administration to beagle dogs at doses of one, three, and 10 gm/kg of body weight. No deaths occurred at any dose. Signs of toxic effect observed at all doses included jumping or struggling during compound administration, tremors and/or subconvulsive movements, defecation, salivation, depression, and ptosis or puffy eyelids. All animals appeared completely recovered within 24 to 48 hours postdose, and appeared normal throughout the remainder of the 28-day observation period. Toxicity was indicated by elevated serum glutamic pyruvic transaminase and serum glutamic oxaloacetic transaminase values, high Juvenile and Band cell counts, depressed hemoglobin and total protein values, and elevated or depressed sedimentation rates in animals given Polyvinylpyrrolidone, with the most marked changes occurring at the high dose (10 gm/kg). All clinical values returned to normal levels shortly after dosing, usually by the 48-hour postdose interval, and remained within normal ranges. (Author)
