The Toxicity of Single Doses of 1-(2-Chloroethyl)-3-(trans-4-Methylcyclohexyl)-1-Nitrosourea (NSC-95,441:MeCCNU) for the Rhesus Monkey (Macaca mulatta) and the Dog.

The report summarizes the results of a study of the toxicity of 1-(2-chloroethyl)-3-(trans-4-methylcyclohexyl)-1-nitrosourea for the rhesus monkey and dog. The compound,designated NSC-95,441,and hereafter as MeCCNU,is the trans-4-methyl derivative of CCNU (NSC-79,037) and a close relative of BCNU (NSC-409,962),two nitrosourea derivatives that are being studied for activity in various neoplastic diseases of man. The data acquired in the major toxicity experiment have shown that MeCCNU produces fatal reactions in the rhesus monkey when administered orally at doses of 200mg per kg and intravenously at doses of 60mg per kg;doses of 50and 30mg per kg are tolerated. In the dog,doses of 25and 12.5mg per kg are regularly fatal when delivered by the oral and intravenous routes,respectively;doses of 12.5and 6.25mg per kg are tolerated.
