Intrathecal Administration of ThioTEPA to Rhesus Monkeys.

ThioTEPA (Lederle) was administered intrathecally by cisternal puncture,twice weekly for four weeks to six rhesus monkeys. Injection caused a syndrome consisting of extensor rigidity with opisthotonos and fasciculation of the facial muscles which lasted 10to 15minutes. This syndrome was also produced by intrathecal administration of a saline solution duplicating the ThioTEPA vehicle but not by Elliot's B solution. Two animals died following three and four doses of 4mg/kg ThioTEPA,two animals died following eight doses of 2mg/kg ThioTEPA,while two animals survived eight doses of 1mg/kg ThioTEPA. Two additional animals survived eight intravenous doses of 1mg/kg ThioTEPA. Toxicity appeared to be systemic with no indication of direct CNS effect due to ThioTEPA. Due to the high and abnormal ionic composition of this formulation,it is not considered suitable for intrathecal administration. (Author)
