Research and Development Study Leading to the Preparation of Reference Reagents for the Cytomegaloviruses.

In order to obtain specific antisera to cytomegaloviruses (CMV),groups of monkeys and goats were immunized with human CMV strains C87and AD169,and simian CMV strain 2757. Periodic bleedings were taken and the animals were bled out two weeks after the final inoculation. The analysis of these sera was made by plaque reduction neutralization tests,in the presence and absense of complement. With both monkey and goat antisera a two-way cross was observed between the two human strains,but there was no interspecies crossing between the human and simian strains. A technique was devised for the concentration and partial purification of large amounts of extracellular virus to be used for the production of antisera relatively free of antibody to host-cell proteins.
