HBAb has been produced in guinea pigs,rabbits,horses,sheep,goats and chimpanzees using a partially purified antigen prepared by (PEG) fractionation. In nonprimates adsorption was performed using PEG at a concentration which precipitated complex but not HBAb or HBAg. PEG can also be used to enhance many immune-precipitant reactions. Immunologically active IgG was found to be present on HBAg prepared by several different methods and tagged with 125I. This provides an explanation for divergent radioimmune assay (RIA) results reported by various laboratories and suggests a mechanism for immune-tolerance in the carrier state. In a double antibody RIA,the first antibody should be non-primate so that the second antibody does not directly precipitate HBAg. A prototype for automatic plasma fractionation has been designed,constructed and proved satisfactory in initial testing. It appears well adapted to specialized fractionation procedures particularly those for obtaining coagulation factors,fibrinogen,Factor VIII and Factor IX,in high yield.
