摘要

Several variables and techniques were studied to determine the optimum conditions for hybridizing Poly I and Poly C molecules. The concentrations of poly I and poly C molecules used in all experiments were a mgm/ml. The variables tested in the technique of hybridization of the homopolymers were solven viz. distilled water and 0.85% sodium chloride,the temperature of hybridization,the time needed for hybridization,the pH of the final preparations of poly I:C complexes,and the presence of phosphate ions in the formulation. The melting properties of the poly I:C complexes were studied. The influence of ribonulease on poly I:C molecules was evaluated with respect to their antiviral and melting properties. The studies done so far indicate that the optimum conditions for hybridizing the homopolymers were addition of solid sodium chloride after mixing the solution of the homopolymers in distilled water,incubation of the mixture of homopolymers for 30minutes at 44C was optimal for hybridization,the pH of the final preparation can be kept at 7.8and,excess phosphate ions were unfavorable for the biological activity of poly I:C preparations. The melting properties of the poly I:C complexes did not influence their antiviral activity in any significant fashion. (Author)
