BCNU, a nitrosourea used for the treatment of various neoplastic diseases,was evaluated for its effects on reproduction, embryonal and fetal development,and perinatal-postnatal behavior and development. Untreated females mated to males given 4or 8mg/kg/wk has significantly few implantations and more resorptions than controls. Discontinuation of treatment for 48days resulted in the recovery of breeding capability. Treatment of female rats with 0, 0.25, 0.75,or 1.5mg/kg/day for 14days prior to breeding and through day 20of gestation caused a dose-dependent decrease in implantations. Doses of 1, 2,or 4mg/kg/day given ip during various 4-day periods of organogenesis or 1.5mg/kg/day given throughout organogenesis were highly teratogenic in the rat with the principal period of sensitivity being on days 6-9. BCNU was not considered to be teratogenic in the rabbit under the conditions of this test.
