Studies to Improve Methods of Platelet Preservation.

The research progress report presents data on short-term platelet storage with preservation of 81% of the initial platelet viability for 72hours of storage in the following variables are strictly followed:room temperature storage 22 C plus or minus 2;constant gentle agitation;storage container made of Fenwal's PL-146plastic;citrate phosphate dextrose (CPD) anticoagulation;and low g forces to prepare the concentrate. Platelet function is also intact as documented by finding an appropriate correction of the bleeding time for the number of circulating platelets in nonrefractory, aplastic thrombocytopenic recipients. Further studies on long-term preservation using methyl sulfoxide (DMSO) as the cryoprotective agent show preservation of 49% of the original viability and normal function.
