Preservation of Mammalian Interferons as Standard Reference Reagents.

The methods for the production,as well as the assay,of crude rabbit interferon stable to pH 2treatment have been defined. The candidate cell cultures suitable for large-scale production of interferon include lung,spleen and kidney;the determination of which of these is the culture of choice for producing large quantities of high-titered interferon will be based on results from experiments now in progress. Of the four likely candidate inducer viruses tested, bluetongue virus induced the highest titers (1,000to 10,000units/ml) without superinduction. Other important factors which have been,or are being evaluated for their influence on interferon production and stability include:the passage level of rabbit cells,the vessel used for cultivation of the cells,the method of inactivation of the inducing virus,and the chemical-physical properties of rabbit interferons produced by different cells,including molecular weight determinations.
