摘要

An assay has been developed for measuring collagen induced serotonin release which is sensitive and stoichiometric. This method allowed a detailed study of conditions and kinetics of the serotonin release reaction. Cleavage of collagen with CNBr and assay of the peptides by the serotonin release reaction allowed the authors to determine with great precision the active sites on the collagen molecule that bind and activate the platelets. Based on the structural information gained from these studies,more than 20peptides were synthesized,some of them possessing significant inhibitory capacity of the release reaction as measured by the labeled serotonin release assay. Analysis of structure activity relationships of these peptides yielded valuable information for defining the structural requirements for recognition and binding to the collagen receptor in the platelet membrane.
