Acute Cardiovascular Effects of Dichloroallyl Lawsone (NSC-126771) Following Constant Infusion to Anesthetized Beaglehounds.

摘要

As a sequel to a study of acute cardiovascular effects of bolus injections of Dichloroallyl lawsone,this study was performed to establish effects following one-hour infusions. ECG's,vascular pressures and resistances,cardiac output,central venous volume,arterial blood gases,and parameters of left ventricular function were measured in anesthetized beaglehounds. A one-hour infusion of Dichloroallyl lawsone in doses of 32.2, 40.0, 46.8and 62.4mg/kg body weight was made,and the above parameters were monitored during 2control periods,and at 20, 40, 60, 80, 100and 120minutes after start of infusion. One dog receiving each of the highest dose levels died. Other changes of significance were:tachycardia, tachypnea,respiratory alkalosis,hyperthermia,increase in central venous volume,diminution of QT and of all voltages to QRS,elevation in pulmonary trunk pressure,and one instance of an intraventricular conduction disturbance. As with the previous Dichloroallyl lawsone study,these changes may be attributable possibly to local medullary acidosis. Increment in central venous volume may be a prodrome of pulmonary edema,and the reduction in QRS voltages deserves further investigation.
