Development and Testing of Pseudointimal-Lined Vascular Devices.

摘要

The development of techniques and characterization of conditions necessary to produce a blood compatible interface are discussed. Despite early problems,some 18cell-lines have been derived from the dog epidermis. Moreover, 27examples of human epidermis were initiated with the successful maintenance of 12separate cell lines. To date,five prostheses have been tested in vivo,one of which was a naked tube and the other four were lined by heterologous dog cells. All five tubes have shown some clotting and current efforts are directed toward defining reasons. Dynamic testing of seeded prostheses in vitro have demonstrated clearly that the cells are tenacious enough to withstand systolic pressures above 200mm Hg and that 5-6000cells per square mm of surface is essential for the development of a smooth pseudointima. Freeze banking of cell lines and seeded prostheses have been successfully undertaken.
