Microbial Contamination of Bovine Serum

Development and application of viral screening methods to detect endogenous bovine virus contaminants in bovine sera used as a medium supplement in live virus vaccine production. Methods included: (1) an ultra-centrifugation concentration technique with passing on bovine cell cultures; (2) technique utilizing monolayers of bovine cell cultures and repeated cell passaging; and (3) utilization of viral interference to detect non-cytopathogenic interferon inducing viruses. Virus screened bovine sera included: (1) Commercial lots of fetal bovine sera from various suppliers; (2) commercially supplied fetal bovine sera from individual fetuses; (3) Newborn calf sera from individual dairy calves less than 72 hours old and (4) Sera from dairy calves in age groups of 3 to 12 months. Additionally, vaccine control fluids supplied by the FDA were screened for bovine viral contamination. The bovine viral contamination rate was found to be highest in fetal bovine sera (33%), followed by newborn calf sera (9%), and calf sera (4%). The bovine viruses were detected in bovine sera and their frequencies identified.
