Cutaneous Irritation in the Topical Application of Methotrexate (NSC No. 740) to New Zealand White Rabbits.

The skin irritation and organ toxicity potential of topically applied Methotrexate (NSC No. 740) was evaluated in New Zealand white rabbits. No skin irritation was noted at concentrations of 1, 3,and 9% of the test compound as defined by the Draize method. This finding was confirmed histopathologically. No treatment-related lesions,either macro-or microscopic,were noted in the kidney,liver,spleen,femur,bone marrow,large or small intestines,and ovaries or testes. These data indicate that topical application of Methotrexate to rabbits produces no observable acute organ toxicity or skin irritation.
