Cutaneous Irritation in the Topical Application of 5-Fluorouracil NSC-19893to New Zealand White Rabbits.

The skin irritation potential of topically applied 5-fluorouracil NSC-19893on New Zealand white rabbits was evaluated utilizing the method of Draize. Histologic studies of selected tissues including skin test sites were performed. Topically applied 5-Fluorouracil at concentrations of 1, 3,and 9percent produced no skin irritation. Histopathologic examination confirmed that the test compound was non-irritating to the skin. No tissue damage was found in the kidney,spleen,liver,large or small intestine,bone,bone marrow,and ovaries or testes of the test animals that could not be ascribed to normal pathogens. It was concluded that 5-Fluorouracil is not a skin irritant and does not produce organ toxicity in rabbits at concentrations of 9% or less when topically applied.
