The distribution and elimination of intravenously administered Methotre-14C (NSC 740) was studied in female, weaning Yorkshire White Pigs. After 7 days an average of 33.66% was excreted via the feces and 15.22% via the urine. The fecal elimination percentage was then used to determine the amount of Methotrexate-14C absorbed after topical application. Among the whole organs sampled following intravenous administration, liver, kidney and ovaries had the highest specific activities of radioactivity. Intestine, bone, skin and fat had the highest specific activity levels of the random tissues sampled. An average of 59.99% of a topically applied dose of Methotrexate-14C was absorbed at the end of 7 days. Significant levels of radioactivity were also detected in skin surrounding the site of application.
