The report is composed of several publications that reflect the diverse nature of research sponsored under this contract. Salient aspects of the reported data reveal that the predominant antigenic determinant of cholera toxin resides in the B subunit and that only antibodies specific for the B subunit are capable of neutralizing cholera toxin. Antibodies against the A subunit are detectable, but do not neutralize cholera toxin. There may be some immunological relationship between the A and B subunit as revealed by radioimmunoassay techniques. Immunization of rabbits with cholera toxoid combined with whole cell vaccine results in a synergistic protective response against line cell challenge of intestinal loops with Vibrio cholerae. Maximum protection was obtained when rabbits were immunized with the toxoid-vaccine combination by both the oral and parenteral routes. Procholeragenoid provided superior protection to any of the formalin or glutaraldehyde toxoids.
