Healthy beaglehounds were anesthetized and instrumented so that various cardiovascular parameters including electrocardiograms,left ventricular pressure and its derivatives,pressures in the pulmonary trunk and aorta,cardiac output,and vascular resistances could be monitored. Two control records were obtained at 15-minute intervals,after which an infusion of Rubidazone was made over a period of one minute. At 5, 20, 40and 60minutes following the end of infusion,records were obtained. Six dogs received the vehicle,while 6dogs in each group were given 2.5, 5.0,or 7.5mg/kg (50, 100,or 150mg/sq. M). All dogs but one survived the 60-minute period of surveillance,and no dog appeared in acute distress. Decreased parameters of left ventricular function paralleled a decrease in left ventricular preload--a Frank-Starling effect. This resulted in decreases in mean pressures within the pulmonary trunk and aorta and increases in vascular resistances. Cardiac output did not change. ECG changes were limited to changes in contour of QRS and ST-T that indicated altered pathways of ventricular activation,secondary T-wave changes,but neither myocardial injury nor dysrhythmia were indicated.
