The metabolism of azo dyes to potentially carcinogenic aromatic amines was studied in hamsters. Male Syrian-golden-hamsters were administered 100 milligrams per kilogram Direct-Black-38 (1937377) or Pigment-Yellow-12 (6358856) intragastrically. Urine samples were collected at intervals for up to 7 days and analyzed for metabolites. Urine from hamsters given Direct-Black-38 contained 206 micrograms (microg) monoacetylbenzidine (3366618), 6.74 microg diacetylbenzidine (30773718), 2.57 microg 4-aminobiphenyl (92671), 3.33 microg benzidine (92875), and 103 microg alkaline hydrolyzable conjugates of benzidine. The author suggests that Pigment-Yellow-12 passes through the gastrointestinal tract of hamsters unchanged. Workers exposed to Direct-Black-38 could be at risk of developing bladder cancer.
