Antisera to Immune Interferons. Annual Report October 1980-September 1981

Hyperimmune anti-globulins to human immune interferon (IFN gamma), mouse IFN gamma and to their respective mock IFNs were produced in rabbits. Rabbits were injected with partially purified preparations of human IFN gamma, mouse IFN gamma or mock IFN preparations once a month. Rabbits were bled twice a month and by the end of one year, 900 ml of pooled sera titering 10,000 units/ml against human IFN gamma and 950 ml of pooled sera titering 6000 units/ml against mouse IFN gamma had been collected. One unit of antiserum neutralizes one unit of IFN. 950 ml of pooled sera from rabbits injected with mock human IFN gamma and 950 ml of pooled sera from rabbits injected with mock mouse IFN gamma were also collected. The ammonium sulphate precipitated globulin fractions of the various pooled antisera were dialyzed against phosphate buffered saline, and then absorbed with cells and antigen bound to Sepharose beads to remove contaminating antibody. Anti-globulin (560 ml) to human IFN gamma had a titer of 60,000 units/ml and antiglobulin (580 ml) to mouse IFN gamma had a titer of 20,000 units/ml. Both were specific for human and mouse IFN gamma respectively. Mock anti-globulin to human IFN gamma (560 ml) and mouse IFN gamma (580 ml) contained no (<10 units/ml) anti-IFN gamma, alpha or beta activity.
