Various doses of N-OH-2-AAF were administered transplacentally to Balb/c mice to determine the highest dose which would not lead to significant mortality in the offspring. It was determined that a dose of 50 mg of N-OH-2-AAF per day on gestational days 16, 17 and 18 was quite toxic to the offspring and led to significant mortality. However, a dose as high as 25 mg per day was well tolerated by the offspring without significant mortality. It was concluded, therefore, that doses of N-OH-2-AAF as high as 25 mg per day on gestational days 16, 17 and 18 would be the preferred treatment in future studies.
