Methods of Producing Human Interferon

The work was concerned with methods of production of two types of human interferon: fibroblast (beta) and immune (gamma) interferon. In addition to the FS-35 cell strain that was selected earlier for fibroblast FS-4 cell strain. These ampules are being stored in liquid nitrogen, available for distribution to qualified investigators by NIAID. The authors have developed a method of production of human IFN-gamma in cultures of mononuclear cells derived from platelet residues that are being made available to us by the New York Blood Center. This method employs combined treatment with phytohemagglutinin (PHA) and 12-0-tetradecanoylphorbol 13-acetate (TPA). This method results in the production of reproducible quantities of IFN-gamma. IFN-gamma produced by this method was purified to a specific activity of 10 to the 7th power units/mg protein. Additional separation by SDS-PAGE resulted in the isolation of two pure subspecies of human IFN-gamma.
