Enhanced Woody Plant Micropropagation through Control of Secondary Phenolic Compounds

Researchers screened a large number of additives to a culture solution thought to be capable of blocking phenolic oxidation or reactivity. Three large-scale studies were carried out: (1) Toxicity testing on previously established cultures of Rhododendron and Nandina; (2) Quantitative aspects of phenolics liberated into additive-containing medium by fresh explants of Bradford Pear, Northern Red Oak, and Cherokee Princess Dogwood; and (3) The ability of these additives to enhance the establishment of micro-propagation cultures of these varieties of Pear, Oak, and Dogwood. While phases (1) and (2) were generally successful, phase (3) was complicated by high levels of endogenous contaminants, the difficulty of obtaining large numbers of explants of a similar size and developmental stage, and the inherently slow growth of young explants. Although the interaction was complicated and often species-specific, the most useful additives included 1 mM of either clutamate, thiourea, or sodium bicarbonate, or 0.1 mM of sodium sulfite.
