Metabolism of Gentian Violet in Chickens

Because of the possibility that gentian violet ingested by poultry may occur as a residue in edible tissues, the metabolism study was undertaken to measure and identify metabolites of gentian violet which may occur in tissues of broiler chickens fed gentian violet at a dosage level commensurate with the recommended level in feed. Broiler chickens of both sexes were dosed with (phenyl-U-14C) gentian violet in amounts duplicating commercial dosing levels for a period of seven days to approach a steady-state residue level of metabolite in edible tissues. Birds were sacrificed at various times after the last dose to provide materials for measurement of depletion of residue. Samples of seven edible tissues were combusted to measure depletion of radiactivity. It was found that depletion of total residue was biphasic in each tissue with graphical half-lives in the range of 59-215 hours for the second phase. Depletion half-lives were consistent between the sexes with the longest half-life of the second phase occurring in liver. Residue equivalents of gentian violet were highest in liver, dropping from 169.1 ppb at 6-hours to 20.9 ppb at 240-hours after the last dose in males. Levels in female liver were approximately half the level in male liver. Residue levels were lowest in breast and thigh at all depletion periods, and dropped to less than 1 ppb after 240-hours.
