Aeration and Agitation in Tissue Culture Using Liquid Carriers

The objective of the research was to demonstrate the technical feasibility of a new concept for increasing the oxygen supply to suspension tissue cultures in order to support high density populations in large scale operations. The concept utilizes a water immiscible, high density liquid with a high oxygen carrying capacity (perfluorocarbon) which is circulated through a draft tube reactor to create bulk mixing. Accomplishments of this study were the demonstration that oxygen transport rates theoretically sufficient to support high culture densities could be achieved and that the perfluorocarbon was non-toxic to the cell line of interest. The concept has yet to be verified with tissue culture cell, as it was found that perfluorocarbon interacts with the serum component of tissue culture medium to form a stable emulsion which disrupted the system.
