Fluorescence Correction in Fermentation Using Evanescent Wave

The study investigated the feasibility of utilizing evanescent wave measurement techniques to measure solution fluorescence in the presence of intracellular fluorescence. Utilizing optical fibers and a flat plate wave guide, a standard fluorescence spectrophotometer was modified to measure evanescent waves emanating from excited cellular suspensions, fluorophore labeled particulates and standard chemical solutions. It was shown that: sensitivity to soluble fluorophores is the same for standard and evanescent wave fluorescence; evanescent wave measurement is much less sensitive to intracellular fluorescence changes than is the standard measurement; when a soluble fluorophore (NADH) is added to a cell suspension, the fluorescences are approximately additive; and leakage from cells is detected by the evanescent wave measurement.
