Selected chemicals were screened for their ability to cause adverse reproductive effects using a postnatal mouse screening test. Attempts were made to determine the median lethal dose to be used and reproductive effects were monitored. Treatments were administered by gavage on gestation days 6 through 15. Probenecid (57669) at 1300 mg/kg/day was toxic to some maternal animals. Benzyl-alcohol (100516) and all trans-retinoic-acid (302794) doses produced no treatment related deaths in maternal animals. Benzyl-alcohol and probenecid caused no significant changes in maternal weight. The predicted median lethal dose for benzyl-alcohol was 550 mg/kg/day and that for probenecid was 1300 mg/kg/day. Neither chemical had a significant effect on the gestation index, reproductive index, maternal status, litter size or postnatal survival, average litter weight or weight gain among the pups. Neither chemical produced any evidence of developmental toxicity despite the maternal mortality caused by probenecid.
