The authors identified a series of N(2-)(dihalophenyl)guanines that were competitive inhibitors of Cytomegalovirus (CMV) DNA polymerase and that inhibited virus-specific DNA synthesis in CMV-infected HEL cells. Assays on the normal host DNA polymerase alpha and on thymidine incorporation by uninfected cells in culture showed that N(2-)(3,5-dichlorophenyl)guanine appeared to be the most selective (least cytotoxic) CMV inhibitor in this series. Complete syntheses and elemental analyses of most of the compounds listed in the first annual report have been done. Several compounds did not give satisfactory analyses after resubmission.
