The primary focus of the Phase I SBIR project was to develop an adjuvant system which is effective in promoting a strong immune response against antigens of low immunogenicity in warm-blooded animals, including man. The approach which was used in this project was based on the known effectiveness of oil-in-water (o/w) emulsions containing monophosphoryl lipid A (MPL) and trehalose dimycolate (TDM) as adjuvants for a variety of antigens. In the project, the feasibility of an adjuvant system in which MPL and TDM are incorporated into liposomes rather than o/w emulsions was examined. It was found that liposomes do provide an effective vehicle for MPL and TDM, and that liposomes containing MPL and TDM are good adjuvants for peptides and proteins, although not for polysaccharides.
