The role of de novo synthesis of acetylcholinesterase (AChE) in the spontaneous recovery of neuromuscular function (NMT) was studied in diaphragms isolated from soman-intoxicated rats. Ten min. after soman injection (3xLD50 i.v.) the AChE activity and NMT appear ed to be fully blocked. AChE activity in endplate and endplate free regions recovered linearly with time (1.5%/h and 2.9%/h, respectively). 180 min. after soman the NMT appeared to be improved. Since both the inhibition of de novo synthesis of AChE by cycloheximide and the repeated inhibition of AChE in vitro did not affect the improvement of NMT, it was concluded that the spontaneous recovery of NMT can not be attributed to synthesis of new AChE.
