The production of specific antibodies to nitropyrenes is desirable for isolation and quantification of nitropyrenes in biological samples from exposed human populations. Dr. Groopman demonstrated the immunogenicity of chemically-modified proteins. He immunized rabbits with 1-nitrosopyrene-modified keyhole limpet hemocyanin that was prepared either by diazotization coupling of 1-aminopyrene to the protein, or by the nitroso condensation of 1-nitrosopyrene to DNA and the carrier protein. The nitropyrene protein antigen, when inoculated into rabbits, generated polyclonal antisera that reacted with 1-acetylaminopyrene, 1-acetylamino-6-nitropyrene, 1-acetylamino-8-nitropyrene, and the C-8-aminopyrene deoxyguanosine-DNA adduct, and 20- to 25-fold less strongly with 1-nitropyrene and 1-nitrosopyrene. The specificity of the antisera has not been proved, however. Unsuccessful attempts to produce specific antisera in mice and to generate monoclonals were also reported.
