Effect of Leucovorin Treatment on Chlorsulfaquinoxaline (NSC-339004) Induced Toxicity in Beagle Dogs

The ability of leucovorin to ameloriate Chlorsulfaquinoxaline-induced toxicity was investigated in Beagle dogs. The dogs were administered a single intravenous dose of Chlorsulfaquinoxaline at 12mg/kg. Beginning one hour after this subcutaneous injections of 10mg/kg were given every 12 hours for a total of 10 doses. Treatment with leucovorin failed to alleviate the toxic effects of Chlorsulfaquinoxaline. All dogs experienced severe gastrointestinal disturbance, significant decreases in body weight and slight to severe leukopenia. All dogs survived to sacrifice on day 29 of the study. Chlorsulfaquinoxaline induced toxicity was partially to completely reversible within the study period.
