The objective of the Phase I research was to scale-up production of hybrid celery somatic embryos in one to four liter bioreactors. researchers established the physical and nutritional conditions necessary for celery embryo acclimation in a bioreactor. The medium was improved, and factors such as temperature, lighting, and culture duration were optimized. Studies of agitation and bioreactor configuration suggested that an airlift bioreactor with a high aeration rate was best suited for production of large numbers of singulated, high quality celery embryos. It was discovered that embryos between 1.0 to 2.5 mm in length produced a greater proportion of normal plants than any other embryo size class. Because the bioreactors have greater capacity than flasks, embryo production in bioreactors was less labor intensive and, thus, more cost efficient.
