Oxygen limitation is a major obstacle in industrial fermentations that utilizeaerobic cell cultures. Antibiotic fermentations using the filamentous bacteria Streptomyces are particularly susceptible to this problem. As a result of oxygen limitation, antibiotic yields in Streptomyces fermentations are often low. The researchers approach to this problem is the development of modified strains that can obtain oxygen from a culture medium more efficiently. A bacterial hemoglobin-like protein was expressed in Streptomyces coelicolor and its effect on various growth parameters and the production of the polyketide antibiotic, antinorhodin, was measured. The hemoglobin was expressed in Streptomyces in a biologically active form as demonstrated by a carbon monoxide binding assay. The expression of functional hemoglobin resulted in a greater than 10-fold increase in the specific yields of actinorhodin.
