Biosensors, based on an affinity reaction between e.g. enzyme/substrate orantibody/antigen, focus interest on surface modification and immobilization techniques. Various electrochemical and optical methods can be used to monitor the interaction either kinetically or at equilibrium. Many of these methods require a stable and reproducible layer of molecules with affinity for the substances to be measured. Furthermore, to obtain appreciable sensitivity and specificity, such aspects as molecular orientation and a nondenaturating environment are important. Stable and reproducible layers of biomolecules such as proteins can be obtained by controlled covalent coupling to a well-defined surface.
