Development of in vitro mutagenicity testing systems using t-lymphocytes

This report presents progress on the following: studies of colon cancer cells with mismatch repair deficiency due to mutations in hMLH1; additional T-cell cloning assays on peripheral blood samples from individuals constitutively heterozygous for hMSH2 or hMLH1 mutations; TCR defined clonality for hprt mutants induced in vitro by low dose low LET ionizing radiations in four independent cultures of T-cells from an individual who is constitutionally heterozygous for a mutation in the ataxia telangiectasia gene; a lymphoblastoid line was initiated from peripheral blood lymphocytes from an individual who is constitutionally heterozygous for an hMLH2 mutation; and studies on children in remission/relapse with acute leukemia.
