Effect of Freund’s Complete Adjuvant Upon Clearance Of Iron-59 Oxide Fron Rat Lungs

On the basis that the number of alveolar macrophages available is a factor in the phagocytic removal of particles from the lungs, the effect of a sustained increase in macrophage concentration, as induced by Freund's complete adjuvant (FCA), upon the clearance kinetics of a test dust (59Fe203) was inves¬tigated. The results of this study in 91 rats showed lung clearance, was increased if the dust was administered 1, 2 or 3 days after FCA treatment, i.e. when the response to FCA was greatest in terms of both number of free macrophages and parenchymal reaction. Both the amount cleared during the first 24 hours and the rate of clearance during the later phase were increased, such that, at 40 days post exposure, FCA-treated rats retained approximately 60％ as much as the controls. Dust clearance was only slightly increased when rats were exposed 7 days after FCA and not affected when exposed either before or 14 days after FCA treatment.
