Preparative Sephadex Column Electrophoresis: An Improved Apparatus

A preparative column electrophoresis using Sephadex G-25 as supporting medium offers a rapid and inexpensive method of purifying a large quantity of proteins with minimum sample loss. This apparatus has been very effective in separating closely related horseradish peroxidase isoenzymes and promises to be of great value in the preparation of other proteins. Somewhat cumbersome but necessary manipulations before and after electrophoresis are major drawbacks to the use of the method. The improved apparatus described simplifies the operation by eliminating exchanges of various plugs, and allows sample loading and elution in situ, without removing the column from buffer reservoirs. Essential parts of the apparatus are fashioned from commercially available items with little modification.
